## In plain language

Colchicine is an old medicine, best known for gout, that has anti-inflammatory effects. In the LoDoCo2 trial, a low daily dose lowered the rate of heart and blood-vessel events in people with chronic coronary disease, though deaths from non-cardiovascular causes were reported at a higher rate with colchicine, with an interval that includes no difference.

## Technical summary

RxNorm ingredient 2683. The LoDoCo2 abstract tested 0.5 mg once daily. The registry record of a separate trial, COLCOT (NCT02551094), lists coronary artery disease and myocardial infarction as conditions and was committed as a source; its results are not recorded here.

## Claims, with the population each applies to

## Context: Patients with chronic coronary disease (LoDoCo2)

Recipient: Adult; Population: Patients with chronic coronary disease; 5522 were randomised (abstract). The abstract does not give an age range. The trial is registered with the Australian New Zealand Clinical Trials Registry, which was not read.; Care setting: Not stated in the source (abstract only); Geography: Not stated in the source (abstract only); Life stage: Adult patients; no age range is stated in the abstract; Follow-up: Median follow-up 28.6 months (abstract).

### In LoDoCo2, 5522 patients with chronic coronary disease were randomised to colchicine 0.5 mg once daily (2762) or placebo (2760). Over a median of 28.6 months, the primary composite end point occurred in 187 patients (6.8%) on colchicine and 264 (9.6%) on placebo (2.5 against 3.6 events per 100 person-years; hazard ratio 0.69, 95% CI 0.57 to 0.83). The key secondary composite occurred in 115 (4.2%) and 157 (5.7%) (hazard ratio 0.72, 95% CI 0.57 to 0.92). Death from noncardiovascular causes was higher with colchicine (0.7 against 0.5 events per 100 person-years; hazard ratio 1.51, 95% CI 0.99 to 2.31).

- Category: Effect
- Evidence stage: Interventional
- Certainty: Certainty not assessed
- Applies to: Recipient: Adult; Population: Patients with chronic coronary disease; 5522 were randomised (abstract).
- Limitations: One randomised, double-blind trial; the abstract names the National Health Medical Research Council of Australia and others as funders. The higher rate of noncardiovascular death is reported as given and its interval includes 1. This record covers LoDoCo2 only. A separate trial of colchicine after a recent heart attack is registered as COLCOT (NCT02551094); this atlas records its registry entry and not its results. Abstract only.
